Abstract

From the iconic Vacanti Mouse of 1997 to today's sophisticated organoid systems, tissue engineering has evolved dramatically. This article traces the field's progression from "top-down" scaffold-based approaches to "bottom-up" developmental biology-inspired methods using iPSCs and CRISPR. The author examines current challenges in achieving hierarchical complexity and scale, arguing that convergence of both approaches will enable the generation of functional, transplantable organs within 25 years.
